are you trying on Windows or Unix.

The Correct Sytax is

http://hostname:portnumber/em

Eg:
http://comp42:5500/em

For getting the Port No:
You can check $ORACLE_HOME/install/portlist.ini file

Refer to the section "Starting the Oracle Enterprise Manager Console" on this link for more details.

http://download-west.oracle.com/docs/cd/B14117_01/server.101/b10742/em_manage.htm#sthref125

Let me know if that works.

Regards
Vamshee.

I got an answer from Oracle on this and it has worked for this problem.
When changing the machine hostname the chgiphost.bat script needs to be run to update all the configuration files
in the ORACLE_HOME so the references to the hostname get updated to the new one
This is documented in the section 8.2.2 of the Administration Guide : http://download-west.oracle.com/docs/cd/B14099_19/core.1012/b13995/host.htm#ASADM026c
Hope this helps somebody else out there a few hours of pain!
