Always recommended for backup when minimum activity on database.

After minimising all the activities, what will be the general time to take a rman backup for 100gb?

It is really very hard to say becuase it is depand on your ORACLE and OS configuration...
of which type of backup >>> full , incremental.

You should try first on test enviourment and after that you implement on production database.

Actual time taken / size of backupsets depends on many factors.
Incremental backup does not essential mean that backups are faster and smaller in size.
It depends the transactions you make. More transactions, more archived logs to backup.
It depdends on your database design. If the tablespace has multiple datafiles spread across, the only datafiles that were active would be backed up.
Depends on channels you have allocated and hardware resources.
