I am using Oracle 10g Release 10.2.0.1 on Oracle Enterprise Linux 4

I have created a few partitions on Linux ( Oracle Enterprise Linux version 4 )

And now I need to create ASM volumes on top of them , so should I create a file system on the new partitions created and then use
/etc/init.d/oracleasm createdisk

(or)

Can i directly run /etc/init.d/oracleasm createdisk on the new partitions which dont contain any file system?

ASM is just a volume manager. Directly run createdisk. ASM will create the filesystem for you.
